Cloning and Analysis of Auxin-binding Protein 1 cDNA Genes in Gossypium hirsutum

Abstract:Auxin binding protein 1(ABP1) plays an important role in plant cell development. Primers were designed coding for conservative amino acid regions in ABP 1 protein family. Using RT-PCR and RACE methods,5′up stream and 3′down stream cDNA sequence of ABP 1 were isolated from Gossypium hirsutum for the first time.Information analysis indicated that the full length cDNA is 824 bp and encodes 190 amino protein. The encoding protein has all of the typical domains of ABP1′s family.It was named GhABP 1.The ABP1 protein′s three residue domains,labelled box A,B and C,are highly conserved among all higher plant ABP 1s. The Box A domain is an important auxin binding site. The WDE acidic residues of the C terminus were shown to play a critical role in protein folding and functional activity of ABP 1 at the plasma membrane. C terminus carries the KDEL motif. The KDEL sequence identifies ABP1 as a protein targeted to and retained in the lumen of the ER. Alignment analysis of the amino sequence deduced from the ABP1 and the sequence of other plants retrieved from GenBank were carried out by Clustal W program , which showed that the identities of the deduced amino acid with Arabidopsis thaliana,Helianthus annuus ,Sinapis arvensis C.annuum,Matricaria recutita are 72%,72%,71%,70% and 70%, respectively.Phylogenetic analysis showed that the amino acid sequence of ABP1 in Gossypium hirsutum first gathered with that in Raphanus sativus,Arabidopsis thaliana and Sinapis arvensis .Sequence analysis indicates that this gene is a new member of the auxin-binding protein 1 gene family.
