Molecular Cloning and Expression Analysis of a Serine/Threonine Protein Kinase Gene in Upland Cotton

Abstract:The cDNA encoding the full length of the serine/threonine protein kinase like protein of cotton was cloned through RT-PCR. The amino acid sequence encoded by it shows an 82% homology to the ATPK3 of Arabidopsis. This gene was tentatively designated as GhPK1 . The entire coding region of GhPK1 is 1038 bp and encodes a polypeptide of 346 amino acids. Its product may possess one trans-membrane domain in the region of 265 to 270 amino acids and may bind to the membrane of the endoplasmic reticulum. GhPK1 shows a higher expression level in seed and fiber than in other organs. In addition, the expression of GhPK1 is increased by salt induction which indicates GhPK1 may participate in salt-stress reactions.
