Prevent Browning of Upland Cotton Embryoid Callus

Abstract：In cotton tissue culture, normal green embryoid is usually obtained after 3 or 4 rounds subcultures. But the callus usually turns brown in the subculture from embryogenic callus to somatic embryogenesis. Our research suggests that the brownturning mainly results from the improper ratio of nitrogen. It also indicated that the content of KT、6-BA and IAA had no significant effect on embryo tissue brownturning. The normal green embryoid is produced only by once subculture after improvement of MS culture medium, shorten its cycle about two months.
