Prevent Browning of Upland Cotton Embryoid Callus Abstract£ºIn cotton tissue culture, normal green embryoid is usually obtained after 3 or 4 rounds subcultures. But the callus usually turns brown in the subculture from embryogenic callus to somatic embryogenesis. Our research suggests that the brownª²turning mainly results from the improper ratio of nitrogen. It also indicated that the content of KT¡¢6-BA and IAA had no significant effect on embryo tissue brownª²turning. The normal green embryoid is produced only by once subculture after improvement of MS culture medium, shorten its cycle about two months. |
