Technical Improvement of Tissue Culture and Somatic Embryogenesis in Upland Cotton

Abstract£ºTwo varieties of upland cotton, Coker 312 and Jiwu 2031, were studied on somatic embryogenesis. They produced different amount of embryogenic calli and regenerated plants. A better procedure of cotton embryogenic callus inducement was established. Special effects of PEG6000 on somatic embryogenesis in upland cotton were investigated. PEG6000 could raise ratio of embryoids germination remarkablely, which got the highest rate of embryoids germination in the treatments of 50 g¡¤L-1 in suspension culture. It promoted embryogenic callus germination in normal light period in two weeks after one or two weeks in darkness with 80-160 mg¡¤L-1 of PEG6000 in solid medium. This method was also applied to maintenance embryonic callus. MSB(MS inorganic salts and B5 vitamin) with phytohormone free, 500 mg¡¤L-1 of CH, half amount of NH4NO4 and double amount of KNO3 could stimulate formation and germination of embryoids, and reduce abnormal embryoids.
