Mapping of RGAP£¬DGAP and DDRT Markers in Cotton

Abstract£ºAccording to the sequences of RGAs and DGAs cloned in our laboratory, 48 primers of RGAP and four primers of DGAP were designed, five RGAP and two DGAP markers were located on the chromosomes of cotton genome using a BC1 population derived from the cross of ¡®Hai 7124¡ÁTM-1'£»the root RNA was extracted in different period after inoculating the isolate of Verticillium dahliae to 5026, a G. hirsutum line high resistant to Verticillium wilt. Totally 164 differential fragments were gotten using mRNA differential display method, and 34 DDRT primers were designed. Three RGAP and five DDRT loci were mapped to the chromosomes using an F2 population derived from the cross of ¡®Hai 7124¡ÁJunmian 1'. The two linkage maps were integrated into one using Joinmap 3.0 software, and the linkage relationship was analyzed between the markers and the QTLs resistant to Verticillium wilt.
